The northbound side of the A56 Dunham Road will be closed for 10 days to allow essential water mains work to be carried out safely.
The closure - which will be put in place on Friday, November 13 - comes as part of United Utilities' £7 million pipe-laying project to improve water supplies for 24,000 customers across the Knutsford and Rostherne areas.
United Utilities' Project Manager Ian Grzelak said: "The work will be carried out in sections along Dunham Road from Dunham Park Service Reservoir through to the south of Charcoal Road.
"In the interests of health and safety we are closing the northbound side of the road for 10 days in agreement with Trafford Highways Department.
"This means that a contraflow system will be in place along the southbound side for all traffic.
"We would therefore advise that motorists plan extra time for their journeys when using this route."
Due to the contraflow on Dunham Road it will not be possible to make a right turn at the Charcoal Road / Park Road crossroads when travelling from any direction.
Those wishing to travel south from Charcoal Road will need to follow the diversion route which will be put in place - taking traffic along Park Road and Bow Green Road.
"We would like to apologise in advance for any inconvenience this closure may cause," added Ian.
"We will be working extended hours and weekends to ensure that we complete this section of work as quickly as possible and have the road reopened by November 22."
The route of the 9.5km pipeline passes close by to Rostherne, Dunham Massey, Tatton Park, Bowden and Knutsford.
The project is expected to be completed during May 2010.
